Detection of TB Bacilli by Polymerase Chain Reaction

Abstract

|
In oder to improve the rate and rapidity in detection of Mycobacterium tuberculosis we evaluated polymerase chain reaction technology using various method. We have compared two DNA isolation methods, 2 polymerase, 3 set of primer and 2 kinds of
thermocycler.
@ES The results obtained are summerized as follows:
@EN 1) In DNA isolation methods, proteinase-K and CTAB method showed equally sensitive, but the latter is more fast.
2) In comparision of three sets of primers 245 base pair fragments of IS986 showed more sensitive result compared to KB5/KB6 primer.
3) In comparision of polymerase, Taq polymerase and Tca polymerase were equally sensitive.
4) In comparision of thermocycler, FTC-2000 was more sensitive and convinient
Our results demonstrate that the modified PCR method is suitable for the rapid detection of tuberculosis in clinical samples. It is recommeded that optimal condition should be used in application to clinical samples.
